Antifungal effects of Pseudomonas aeruginosa MB1-3 and its active compound against fish pathogenic Saprolegnia sp.

Abstract
The aim of this study was to evaluate the possibility of controlling saprolegniosis of rainbow trout eggs using an ethyl acetate extract of culture supernatant of Pseudomonas aeruginosa MB1-3 (MB-EA). Antifungal activity of MB-EA against Saprolegnia sp. FP9001 was determined. All experimental groups treated with MB-EA at different concentrations showed significant (all p < 0.05) inhibition of mycelial growth compared to the control group. At 50 mg/L and 500 mg/L, MB-EA reduced hyphal growth by 18.3% and 55.9%, respectively.
Spore germination was evaluated after treatment with MB-EA at various concentrations. At 50 mg/L and 1000 mg/L, MB-EA inhibited fungal spore germination by 24.3 ¡¾ 3.2% and 65.0 ¡¾ 10.8%, respectively. Toxicities of MB-EA to CHSE-214 and RTG-2 fish cell line were evaluated using MTT assay. When cells were treated with MB-EA at concentration up to 250 mg/L, there was no significant difference in cell viability from the control group. Moreover, survival rates of fertilized eggs and hatching rates of eyed eggs of rainbow trout did not differ between control group and the group treated with MB-EA at 30 min/day for 2 days. An active compound was isolated from MB-EA by RP-MPLC and HPLC. Its chemical structure was identified as 2-heptyl-4-quinolone (HHQ) through MS, 1H-NMR, and 13C-NMR analyses. This active compound exhibited antifungal activity with a minimum inhibitory concentration (MIC) value of 200 mg/L. Therefore, MB-EA and its active compound HHQ exhibit antifungal activity and they can be used as candidates for controlling saprolegniosis of fertilized eggs in rainbow trout farms.
